Vliv věku jedince na xylogenezi smrku ztepilého (Picea abies (L.) Karst.) během vegetačního období

In this work, it was analyzed whether the age of the tree has an influence on the radial growth in stem. Samples were taken from trees of different ages during the growing season at Rájec-Němčice research site in 2017. Paraffin blocks were made from the samples to cut the micro-incisions and then permanent microscopic preparations were made. Cambium and xylem cells produced last year were observed using a microscope. Cambial activity was 21 days longer in young trees. Maximum cell production was at the end of June. The average total increment was 49 cells in young trees and 38 cells in old trees.

The content of this diploma thesis is the analysis of the formation of beech wood (Fagus sylvatica L.) at the cellular level, ie xylogenesis in the research area Rájec - Holíkov, which is located in the Drahanská Highlands. Specifically, this study focused on the activity of cells in the cambial zone and the differentiation of the new annual ring on individual days (DOY) during the 2016 growing season compared to 2015, when samples were taken (Trephor tool) regularly once a week using so-called micro-bore. Permanent microscopic slides were created from these, which were subsequently examined. The activity of cells in the cambial zone was started in 2015 and 2016 in the last week of April at a temperature of 8.3 ° C. The zone of secondary cell wall (SW) formation and gradual cell lignification was evident two weeks after the onset of postcambial growth until the end of August. After the application of the Gompertz function, the creation of a new annual ring took an average of 91 days in 2015 and its size was 1252.7 μm. The year 2016 was similar, the size was 1229.2 μm. The average time of creation of a new annual ring was higher, namely 117 days.
Xylogeneze buku lesního (Fagus sylvatica L.) na Drahanské vrchovině v průběhu dvou po sobě jdoucích vegetačních období

Štěpán Sýkora xylogenetics off European beech (Fagus sylvatica L.) on location Drahanská vrchovina. Bachelor thesis deal with cambial acitvity and the proces of cell differentiation of beech during two consecutive growing seasons according to weather conditions. On the research area Rájec Němčice in 2014 there was carried out on three sample trees. Sampling took place over a weekly period from 13.3. to 20.11. 2014. From taken samples were developed sustained microscopic preparations, from wich the data were obtained by using a light microscope. Monitored were the beginning, the maximum of produciton and the decrease of cambial aktivity. On the new cells there was observed the proces of the cell diferentiation from phase of radial expansion to ending of growth ring production. The obtained data were interleaved by applying the Gompertz function and the individual maximum radial additions and the period of creation of the growth ring and its parts were determined. This bachelor thesis is focused on study to xylogenesis, what analyse a dynamic of growth of xylem and phloem development of Norway spruce (Picea abies (L.) Karst). For this analysis was used methods od radial growth (cambial growth). Samples were taken by from six sample trees in Rajec-Němčice (Drahanská upland). This locality stands between 610 to 625 metres above sea level. Samples for this work were taken every week from 5.4. to 15.11. Permanent mounts were taken as a microbores, which were evaluated by microscope and by program called ImageJ. Cambial activity started at all trees similarly. It started on April. This month were recorded sufficiently high temperature to start a cambial activity. New cells were created in average 0,15 cells at a young trees and 0,11 cells at an old sample trees.

The analysis of secondary growth of xylem was carried out during the growth season in 2017 in the research surface of Rájec-Němčice. The work was focused on a cambial activity and a differentiation of wood cells of two different age stands. Samples were taken by Trephor tool at weekly intervals. Subsequently, permanent preparations were produced from them and microscopically evaluated. Cambial cells were active from the end of April to August. Younger trees had a longer cambial activity per week. Tracheid cells of older trees appeared one week earlier than tracheid cells of younger trees. The trees showed a balanced increase in both cell number and increment size.
MICROCLIMATIC INFLUENCES ON WOOD PHENOLOGY OF TREES AND SHRUBS IN TREELINE ECOTONE

This thesis deals with the influence of microclimatic conditions (air temperature near tree/shrub top, temperature of the stem and root zone) on the wood phenology of trees (Picea abies) and shrubs (Pinus mugo) growing on the same site in the alpine tree line (ATL) ecotone. Phenological and microclimatic research was carried out in the Giant Mountains at 1,370 m a. s. l. in the 2017 vegetation period, with the aim to compare the course of wood formation depending on the microclimate, to determine the temperature thresholds of wood growth and differences in the morphology of trees and shrubs. Based on the microclimate analysis of trees and shrubs, individual variables were determined (average, average minimum, average maximum, daily amplitude of air, stem and root zone temperatures) characterizing the growing season. The collection of micro-drills of trees and shrubs took place from April to October in an interval of 6 - 11 days, which were subsequently processed and examined in the laboratory. The following phenophases of wood development were distinguished: cambial, enlarging, wall-thickening, mature cells and cells of the previous ring.
